DIversité - Adaptation - DEveloppement des plantes

Members and partners of UMR DIADE have access to a fully equipped laboratory dedicated to plant sample preparation and imaging. Various protocols for fixation, clearing, embedding, sectioning, and staining are available, including for complex materials such as lignified tissues or fruits, and for sensitive experiments such as in situ hybridization.

The facility is equipped with a Microm HM650 V vibratome, two Microm HM355 S microtomes for paraffin or resin-embedded material, a Leica JUNG RM 2055 microtome, and a Reichert ultracut S ultramicrotome.

Sample imaging can be performed using a Nikon SMZ1500 binocular microscope equipped with an X-cite emission source and GFP, RFP, CFP and YFP fluorescence filter sets; two Leica DMRB and ARISTOPLAN epifluorescence microscopes with DIC optics; a Zeiss Axio Imager A1 epifluorescence microscope associated with a Colibri 4-LED emission system, and a recent Zeiss Axio Imager Z2 epifluorescence microscope coupled to an X-cite source.

Images can be acquired and processed using digital cameras and computer stations equipped with the Q-capture Pro software or the Zeiss ZEN software.

Various systems of imaging (confocal imaging, light sheet microscopy…) are also available in the vicinity of the IRD centre thanks to the Montpellier RIO Imaging network.
